RB1基因编码肿瘤抑制蛋白Rb,Rb可通过在细胞做好分裂准备之前抑制细胞周期进程来阻止过度的细胞生长。如果在生命早期发生RB1双等位基因突变,Rb蛋白失活,会导致视网膜母细胞瘤形成。 将loxp位点插入到exon3区域两侧,构建Rb1基因条件性敲除小鼠模型。可与组织特异性Cre工具鼠交配,获得在特定细胞类型中敲除Rb1基因的小鼠模型。用于研究Rb1在发育和肿瘤发生中所起作用,以及癌症或细胞周期、衰老研究。Rb1基因敲除纯合子小鼠由于肝脏中不能产生红细胞而在胚胎期死亡,杂合子敲除小鼠在8个月大时发生垂体瘤。
