应用领域:属于化学遗传学元件,用于神经科学研究,药物(叠氮平-N-氧化物,CNO)诱导后GiDREADD(hM4Di)可抑制神经元的活性。hM4Di是一种突变的GPCR,可以激活Gi调节的信号通路,抑制神经元的放电活动。也有研究表明,hM4Di可以抑制神经递质的释放,从而达到抑制神经元活动的效果。

Fig1.荧光检测Dppa3-Cre+/-;R26-hM4Di-2A-mCitrine+/- 小鼠脑内mCitrine的表达。

Dppa3-Cre+/-小鼠与R26-hM4Di-2A-mCitrine+/- 小鼠进行交配获得的双阳性小鼠,取脑组织进行荧光显微镜拍照。实验结果显示R26-hM4Di-2A-mCitrine小鼠可以在脑组织神经元中正确表达。
