Fig.1 Detection of GIPR expression in brain by RT-PCR. Brain RNA was extracted from 12-14-week-old female wild-type C57BL/6 mice (WT) and homozygous hGIPR knockin mice (HO), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human mRNA primers. Mouse Gipr mRNA (177 bp) was detectable only in wild-type C57BL/6 mice. Human GIPR mRNA (343 bp) was detectable only in homozygous hGIPR knockin mice but not in wild-type mice.

Fig.2 Detection of GIPR protein expression in hGIPR mice by WB. The pancreas, brain, liver and brown adipocyte tissue (BAT) lysates were collected from 12-14-week-old female wild-type C57BL/6 mice and homozygous hGIPR mice, and then analyzed by western blot with anti-GIPR antibody. GIPR was obviously detectable in pancreas and liver from both WT C57BL/6 and homozygous hGIPR mice, as the antibody was cross-reactived between human and mouse GIPR.
