Thymus RNA was extracted from male 8-10-week-old wild-type C57BL/6 mice (WT) and heterozygous hCCR9 knockin mice (HE), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human mRNA primers. Mouse Ccr9 mRNA (188 bp) was detectable in all mice. Human CCR9 mRNA (164 bp) was detectable only in heterozygous hCCR9 knockin mice but not in wild-type mice.

Leukocytes were collected from thymus and spleen of wild-type C57BL/6 mice (male, 8 weeks) and heterozygous hCCR9 mice (male, 10 weeks), and analyzed by flow cytometry with anti-mCCR9 and anti-hCCR9. mCCR9 were detectable in all mice. hCCR9 were exclusively detectable in hCCR9 humanized mice but not in wild-type mice.
