Fig.1 Detection of PTGER2 expression in hippocampus, lung, spleen, liver and brain by RT-PCR. Mouse Ptger2 mRNA (227 bp) was detectable only in wild-type C57BL/6 mice. Human PTGER2 mRNA (358 bp) was detectable only in homozygous hPTGER2 knockin mice but not in wild-type mice. Hippocampus, lung, spleen, liver and brain RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=2) and homozygous hPTGER2 knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

PTGER2 was detectable in liver, lung and spleen from both WT C57BL/6 and HO hPTGER2 mice, as the antibody was cross-reactived between human and mouse PTGER2. The liver, lung and spleen lysates were collected from 6-week-old male wild-type C57BL/6 mice and 6-week-old male homozygous hPTGER2 mice, and then analyzed by western blot with anti-PTGER2 antibody.
