Fig.1 Detection of TARDBP expression in frontal lobe by RT-PCR. Mouse Tardbp mRNA (174 bp) was detectable only in wild-type C57BL/6 mice. Human TARDBP mRNA (449 bp) was detectable only in homozygous hTARDBP-A315T knockin mice but not in wild-type mice. Frontal lobe RNA was extracted from 7-year-old male wild-type C57BL/6 (WT)(n=3) and female homozygous hTARDBP-A315T knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.3 Detection of hTARDBP expression in hTARDBP-A315T mice by WB. Human TARDBP was robustly expressed in the brain and spinal cord of homozygous hTARDBP-A315T mice. Lysates of brain and spinal cord tissues were collected from 7-week-old female wild-type C57BL/6 mice and 7-week-old female homozygous hTARDBP-A315T mice, then analyzed by western blot with anti-hTARDBP antibody .

Abbr. HO, homozygous; WT, wild type.

Fig.4 Detection of hTARDBP expression in hTARDBP-A315T mice by WB. Significant overexpression of TARDBP was observed in the brain and spinal cord of homozygous hTARDBP-A315T mice relative to WT C57BL/6 mice, as measured by an antibody cross-reactive with both human and mouse TARDBP. The brain and spinal cord tissue lysates were collected from 7-week-old female wild-type C57BL/6 mice and 7-week-old female homozygous hTARDBP-A315T mice, and then analyzed by western blot.

Abbr. HO, homozygous; WT, wild type; PC, positive control, Hela cells.
