Lung RNA was extracted from wild-type C57BL/6 mice (WT) and homozygous hNOTCH3 knockin mice (HO), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human Notch3 primers (n=1-3, female, 10 weeks old). Mouse Notch3 mRNA (255 bp) was detectable only in wild-type C57BL/6 mice. Human NOTCH3 mRNA (141 bp) was detectable only in homozygous hNOTCH3 knockin mice but not in wild-type mice.

The lung and mammary gland tissue lysates were collected from 10-week-old female wild-type C57BL/6J mice (WT) and 10-week-old female homozygous hNOTCH3 mice (HO), and then analyzed by western blot with anti-NOTCH3 antibody (CST, 5276T). 

The antibody is cross-reactive between human and mouse NOTCH3. PC, positive control, MCF7 cells.
