Fig.1 Detection of mouse IL3 expression in splenocytes of WT C57BL/6 mice and HO IL3-KO mice by ELISA. Mouse IL3 was exclusively detectable in wild-type C57BL/6 mice. Splenocytes were isolated from male 7-week-old wild-type (WT) and male 7-week-old homozygous IL3-KO mice, then seeded in 6-well plates at a density of 3×106 cells/well in 3 mL culture medium. Following in vitro stimulation with 5 μg/mL conA antibody and 1 ng/mL anti-mouse IL2 antibody for 72 h, mouse IL3 levels in the culture supernatants were measured using a anti-mIL3 ELISA kit.
