Fig.1 Detection of TACR2 expression in adrenal by RT-PCR. Mouse Tacr2 mRNA (163 bp) was detectable only in wild-type C57BL/6 mice. Human TACR2 mRNA (221 bp) was detectable only in homozygous hTACR2 knockin mice but not in wild-type mice. Adrenal RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=2) and homozygous hTACR2 knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR.

Fig.2 Detection of TACR2 expression in hTACR2 mice by WB. TACR2 was detectable in colon and trachea from both WT C57BL/6 and HO hTACR2 mice , as the antibody was cross-reactived between human and mouse TACR2. The colon and trachea tissue lysates were collected from 6-week-old male wild-type C57BL/6 mice and homozygous hTACR2 mice, and then analyzed by western blot.

The expression levels of mouse Neurokinin A (NKA) in small intestine homogenates were similar between WT C57BL/6 and homozygous hTACR2 knockin mice. Small intestine homogenate were collected from wild-type C57BL/6 mice (n=2, male, 6 weeks old) and HO hTACR2 mice (n=2, male, 6 weeks old), and analyzed by ELISA.
