Cerebellum, cortex, frontal lobe and lung RNA were extracted from 6-week-old male wild-type C57BL/6 (WT)(n=3) and heterozygous hAPP(3) knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse App mRNA (319 bp) was detectable in both wild-type C57BL/6 mice and heterozygous hAPP(3) knockin mice. Human APP mRNA (282 bp) was detectable only in heterozygous hAPP(3) knockin mice but not in wild-type mice.

The hippocampus, cortex, liver and cerebellum tissue lysates were collected from 6-week-old male wild-type C57BL/6 mice and 6-week-old male heterozygous hAPP(3) mice, and then analyzed by western blot with anti-APP antibody. APP was detectable in hippocampus, cortex, liver and cerebellum from both WT C57BL/6 and HE hAPP(3) mice , as the antibody was cross-reactived between human and mouse APP.

APP was detectable obviously in cortex, hippocampus and cerebellum while not obvious in liver from HO hAPP(3) mice with a human APP-specific antibody. The cortex, hippocampus, cerebellum and liver tissue lysates were collected from 6-week-old male wild-type C57BL/6 mice and 6-week-old male homozygous hAPP(3) mice, and then analyzed by western blot.
