Tissue RNA was extracted from female 8-week-old wild-type C57BL/6 mice (WT) and homozygous hPLA2G10 knockin mice (HO), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human mRNA primers. Mouse Pla2g10 mRNA (209 bp) was detectable only in wild-type C57BL/6 mice. Human PLA2G10 mRNA (210 bp) was detectable only in homozygous hPLA2G10 knockin mice but not in wild-type mice.

Tissues homogenates of the pancreas and lung were collected from wild-type C57BL/6 mice (WT) and homozygous hPLA2G10 knockin mice (HO) (n=2, female, 8 weeks old). Protein expression level of PLA2G10 was analyzed by ELISA. The ELISA kit is cross-reactive between human and mouse PLA2G10 protein.

Tissues lysates of kidney, liver, lung, pancreas and colon were collected from female 8-week-old wild-type C57BL/6J mice (WT) and homozygous hPLA2G10 knockin mice (HO), and then analyzed by western blot with anti-PLA2G10 antibody (Proteintech, 19502-1-AP). 20 μg of total proteins were loaded for western blotting analysis. The antibody is cross-reactive between human and mouse PLA2G10. The positive control (PC) used is MCF-7 cells.
