将CreERT2-Wpre-polyA插入到小鼠Gli1基因起始密码子处。 Gli1是锌指蛋白Kruppel家族的成员。编码的转录因子由sonic hedgehog信号转导级联激活,并调节干细胞的增殖。该蛋白的活性和核定位受到p53的负向调节,形成一个抑制性循环。当这些Gli1-CreERT2小鼠与含有感兴趣的loxP侧翼序列的小鼠交配时,他莫昔芬诱导的Cre介导的重组将导致Gli1表达细胞中侧翼序列的缺失;使它们有助于研究胚胎发育的不同阶段刺猬反应细胞的轴型、增殖和细胞命运规范。
