将CreERT2敲入小鼠内源Isl1(ISL1 transcription factor, LIM/homeodomain )基因ATG位置,构建可受Tamoxifen诱导的Isl1-Cre工具鼠。利用该小鼠,可在心脏和后肢祖细胞中敲除flox的目的基因。该品系杂合子小鼠可正常存活繁育;纯合子小鼠Isl1缺失会引发胚胎致死。

图1 Isl1CreERT2/+;R26tdtomato/+ 胚胎的心脏中可检测到tdTomato的表达。

在Isl1CreERT2/+;R26tdtomato/+双转基因鼠E8.0时注射tomoxifen,可在E13.5的双转基因鼠心脏中检测到tdTomato的表达。Isl+细胞主要分化为右心室细胞。(见参考文献)
