Fig.1 Detection of KLRC1 expression in spleen by RT-PCR. Tissue RNA was extracted from WT C57BL/6 litters and HO hKLRC1 knockin mice (n=2, female, 8 weeks old), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Human KLRC1 mRNA (168 bp) was detectable only in HO hKLRC1 knockin mice but not in WT mice. Mouse KLRC1 mRNA (247 bp) was detectable only in WT C57BL/6 mice.

Leukocytes were collected from spleen and blood of wild-type C57BL/6 mice (female, 8 weeks) and homozygous hKLRC1 mice (female, 8 weeks), analyzed by flow cytometry with anti-mKLRC1 and anti-hKLRC1. hKLRC1 were exclusively detectable in hKLRC1 humanized mice but not in wild-type mice.
